    RPHPLC Determination of nor-CClT the Precursor in Preparation of the Labelled Compound 18F-FECNT

    • A method of RP-HPLC was proposed for the determination of nor-CClT or in its whole chemical name,2β-carbomethoxy-3β-(4-chlorophenyl) nortropane which is the precursor in preparation of the labelled compound,18F-FECNT,or in its whole chemical name Fluoro 18F-N-2-fluoroethyl-2β-carbomethoxy-3β-(4-chlorophenyl) nortropane. The Lichrospher C18 column was used as stationary phase,a mixture of methanol,water and triethylamine mixed in the ratio of 65 to 35 to 0.21 (by vol.) was used as mobile phase and UV-detection at the wavelength of 230 nm was adopted in the determination. Under the optimum condition,clear separation of nor-CClT from the co-existing impurities was attained. Linear relationship between values of peak area and mass concentration of nor-CClT was obtained in the range of 0.100 to 1.200 g·L-1. Values of intraday RSD′s (n=5) found by testing at 3 concentration levels of 0.400,0.600,1.000 g·L-1 were 0.53%,0.46% and 0.32% respectively.
