HPLC Determination of Contents of Quercetin and Kaempferol in Different Positions of Impatiens Balsamina Plant

Abstract
Contents of quercetin and kaempferol,as the active medicative components of Impatiens balsamina in different positions of its plant were determined by HPLC and a comparative study was made.The DiamonisilTM C18 (4.6 mm×250 mm,5 μm) column was used stationary phase,and a mixture of methanol and dil.acetic acid (0.5+99.5) mixed in the ratio of 65 to 35 (pH 3.5) was used a mobile phase with a flow-rate of 0.9 mL·min-1.UV-detection was made at the wavelength of 365 nm.Linear relationship between values of peak area and concentration was obtained in the range of 0.84-6.72 mg·L-1 for quercetin,and in the range of 2.56-20.48 mg·L-1 for kaempferol.Tests of recovery were made by standard addition method,and values of average recovery found were 101.1% and 96.0% with RSD′s (n=5) of 1.5% and 2.6% respectively.It was shown that highest content of quercetin (0.112 mg·g-1) was found in the leaves before blossoming and highest content of kaempferol (0.972 mg·g-1) was found in the flower.
