RPHPLC Determination of 5 Water Soluble Components in Sliced Roots of Salvia Miltiorrhiza with Diode Array Detection

Abstract
RPHPLC was applied to the determination of 5 water soluble components,namely,danshensu,protocatechuic acid,protocatechuic aldehyde,rosmarinic acid and salvianolic acid B,in the TCM of sliced roots of Salvia miltiorrhiza.The sample was extracted with water and the extract was used for the determination.DiamonsilTM C18 column (250 mm×4.6 mm,5 μm) was used as stationary phase with column temperature at 30 ℃,and mixtures of methanol and dil.acetic acid (5+95) in different ratios were used as mobile phase in gradient elution with flow-rate of 1.0 mL·min-1.Wavelength of 298 nm was chosen for UV-detection of the 5 components with the DAD.Well separation was attained and linearity ranges were obtained between 27.4-191.8 mg·L-1 (for danshensu),0.742-5.194 mg·L-1 (for protocatechuic acid),0.526 5-3.683 mg·L-1 (for protocatechuic aldehyde),2.268-15.88 mg·L-1 (for rosmarinic acid),7.728-54.10 mg·L-1 (for salvianolic acid B).Values of average recovery found were 101.5%,100.0%,100.3%,101.0%,102.3% respectively.Significant differences among the contents of the 5 components were found in samples grown in different areas in our country as shown by the analytical results obtained by this method.
