    On the Interaction of Amikacin with Compound Complex of CdTe Quantum Dots and Bovine Serum Albumin

    • The CdTe quantum dots (abbreviated as CdTe-QD′s),which behaved to possess excellent optical characteristics,was synthesized by the hydrothermal method from CdCl2 solution and NaHTe solution using mercaptopropionic acid as stabilizer and poly-N-vinyl-2-pyrrolidone as surfactant.It was found that the intensity of fluorescence of the CdTe-QD′s was increased significantly when the CdTe-QD′s were reacted with bovine serum albumin (BSA) to form a compound complex.Linear relationship between the magnitude of incerase in fluorescence intensity and concentration of BSA was obtained in the range of 0.68-13.6 mg·L-1 with detection limit (3S) of 0.26 mg·L-1.It was found also that through its interaction with amikacin,fluorescence of the compound complex CdTe-QD′s-BSA was remarkably quenched,and the magnitude of decrease of fluorescence intensity was found to keep a linear relationship with the increase in concentration of amikacin in the range of 2.1-64 mg·L-1.It was deduced from the experimental results that the reaction mechanism was a static quenching process,in which amikacin bound with BSA in the mole ratio of 1 to 1.The binding constant found was 6.47×103L·mol-1.Circular dichroism spectrometry was used in the study of the effect of amikacin on the configuration of BSA.
