HPLC Determination of Water Soluble Saccharides in Tobacco

Abstract
A method of HPLC for the determination of water soluble saccharides in tobacco was proposed. The sample was extracted vortically with 0.01 mol·L-1 NaOH solution for 60 min, and the extract obtained was separated by isogradient elution with pure water on Metacarb 67C column (300 mm×6.5 mm), and determined with differential refraction detector. The main water soluble saccharides in tobacco were successfully separated within 12 min, and detection limits (3S/N) found were 0.60 mg·L-1 for fructose, 0.71 mg·L-1 for glucose and 0.39 mg·L-1 for sucrose. Values of recovery and RSD′s (n=8) of the 3 water soluble saccharides found were in ranges of 98.1%-99.2% and 0.97%-1.6% respectively.
